Este o imunitate cu care ne na\u015ftem, este rapid\u0103, nespecific\u0103, f\u0103r\u0103 memorie, independent\u0103 de natura antigenului.
Principalul mecanism de ac\u0163iune, const\u0103 \u00een expresia pe suprafata unor celule (keratinocite, celule NK, celule dendridice, monocite, macrofage), a unei familii de proteine transmembranare (toll like receptor), care recunoa\u015fte un patern molecular de pe suprafa\u0163a agentului patogen microbian. Activarea acestor receptori induce un semnal de transduc\u0163ie care produce activarea factorilor de transcrip\u0163ie (NFkB) cu producerea de TNF, IL1, IFN\u03b3 \u015fi ROS. Aceste citokine sunt implicate direct sau indirect in distrugerea agentului patogen, activarea celuleor inflamatorii, inducerea unei remanieri tisulare, mobilizarea \u015fi activarea celulelor componente ale r\u0103spunsului\/ sistemului imun adaptativ.
Keratinocitele, celulele NK, celulele dendridice, monocitele\/macrofagele exprim\u0103 pe suprafa\u0163a lor proteine transmembranare, membre ale familiei TLR, astfel aceste celule particip\u0103 at\u00e2t la distrugerea germenilor patogeni c\u00e2t \u015fi la celelalte procese, ca inflama\u0163ia, activarea imunit\u0103\u0163ii specifice, care stau la baza unor afec\u0163iuni precum lupus eritematos, pemfigus vulgar, psoriazis.
\u00cen patogenia lupusului eritematos sistemic s-a eviden\u0163iat implicarea masiv\u0103 a imunit\u0103\u0163ii \u00eennascute, astfel at\u00e2t acizii nucleici rezulta\u0163i din distruc\u0163ia celulara, c\u00e2t \u015fi complexele imune circulante pot activa receptroii TLR (TLR7 \u015fi TLR9) care se g\u0103sesc exprima\u0163i \u00een exces pe celulele mononucleate din sange. Innate immunity in autoimmune and inflammatory skin diseases<\/span>

It is the immunity one is born with, it is fast, non-specific, without memory and independent of the nature of the antigen. The main mechanism of action consists in the expression on the surface of several cells (keratinocytes, NK cells, dendritic cells, monocytes, macrophages) of a toll-like protein family that recognize a molecular pattern on the surface of a microbial pathogen. Activation of these receptors induces a transduction signal that produces the activation of transcription factors (NFkB) with the production of TNF, IL1, IFN\u03b3 and ROS. These cytokines are directly or indirectly involved in pathogen destruction, inflammatory cell activation, induction of tissue restoration, mobilization and activation of the constituent cells of the adaptive immune system response.
Keratinocytes, NK cells, dendritic cells, monocytes, macrophages express transmembrane proteins on their surface, which are members of the TLR family, so these cells participate in both the destruction of pathogenic germs and other processes, such as inflammation, activation of specific immunity, being the basis for diseases such as lupus erythematosus, pemphigus vulgaris, psoriasis.
In the pathogenesis of systemic lupus erythematosus, the massive implication of innate immunity has been demonstrated, so both nucleic acids resulting from cellular destruction and circulating immune complexes can activate TLR receptors (TLR7 and TLR9) that are found to be excessively expressed on mononuclear cells in the blood. Also, TLR 7 and TLR9 are involved in autoantibody production, and TLR 2 and TLR 4 are involved in regulating the activity of dendritic cells and Th17 lymphocytes in IL17 production. In pemphigus vulgaris, an increase in TLR4 expression in keratinocytes was seen in both the basal layer and the upper epidermal layers. In psoriasis, keratinocytes excessively express TLR1, TLR2 and TLR5, with significant differences between normal skin and lesional skin. The keratinocyte from injured skin, TLR receptor-activated (TLR2, TLR3, TLR4) increases the production of TNF alpha, IL8 and other proinflammatory cytokines. Activation of innate and adaptive immune response leads to the activation of T ymphocytes, Th1 and especially Th17 lymphocytes, with excess IL17 production. Excessive proinflammatory cytokines as well as the action of LL37 peptide induces excessive proliferation of keratinocytes.
Highlighting the implication of innate immunity in cutaneous pathology opens new perspectives on understanding the pathogenic processes that underlie several types of skin disorders, especially in the initiation phase of these processes.<\/p>\n

Keratinocytes, NK cells, dendritic cells, monocytes, macrophages express transmembrane proteins on their surface, which are members of the TLR family, so these cells participate in both the destruction of pathogenic germs and other processes, such as inflammation, activation of specific immunity, being the basis for diseases such as lupus erythematosus, pemphigus vulgaris, psoriasis.
In the pathogenesis of systemic lupus erythematosus, the massive implication of innate immunity has been demonstrated, so both nucleic acids resulting from cellular destruction and circulating immune complexes can activate TLR receptors (TLR7 and TLR9) that are found to be excessively expressed on mononuclear cells in the blood. Also, TLR 7 and TLR9 are involved in autoantibody production, and TLR 2 and TLR 4 are involved in regulating the activity of dendritic cells and Th17 lymphocytes in IL17 production. In pemphigus vulgaris, an increase in TLR4 expression in keratinocytes was seen in both the basal layer and the upper epidermal layers. In psoriasis, keratinocytes excessively express TLR1, TLR2 and TLR5, with significant differences between normal skin and lesional skin. The keratinocyte from injured skin, TLR receptor-activated (TLR2, TLR3, TLR4) increases the production of TNF alpha, IL8 and other proinflammatory cytokines. Activation of innate and adaptive immune response leads to the activation of T ymphocytes, Th1 and especially Th17 lymphocytes, with excess IL17 production. Excessive proinflammatory cytokines as well as the action of LL37 peptide induces excessive proliferation of keratinocytes.
Highlighting the implication of innate immunity in cutaneous pathology opens new perspectives on understanding the pathogenic processes that underlie several types of skin disorders, especially in the initiation phase of these processes.<\/p>\n

Imunitatea \u00eenn\u0103scut\u0103 constituie una din cele doua laturi ale imunit\u0103\u0163ii generale, fiind totodat\u0103 prima linie de aparare \u00eempotriva agen\u0163ilor patogeni. Este o imunitate cu care ne na\u015ftem, este rapid\u0103, nespecific\u0103, f\u0103r\u0103 memorie, independent\u0103 de natura antigenului.
Principalul mecanism de ac\u0163iune, const\u0103 \u00een expresia pe suprafata unor celule (keratinocite, celule NK, celule dendridice, monocite, macrofage), a unei familii de proteine transmembranare (toll like receptor), care recunoa\u015fte un patern molecular de pe suprafa\u0163a agentului patogen microbian. Activarea acestor receptori induce un semnal de transduc\u0163ie care produce activarea factorilor de transcrip\u0163ie (NFkB) cu producerea de TNF, IL1, IFN\u03b3 \u015fi ROS. Aceste citokine sunt implicate direct sau indirect in distrugerea agentului patogen, activarea celuleor inflamatorii, inducerea unei remanieri tisulare, mobilizarea \u015fi activarea celulelor componente ale r\u0103spunsului\/ sistemului imun adaptativ.
Keratinocitele, celulele NK, celulele dendridice, monocitele\/macrofagele exprim\u0103 pe suprafa\u0163a lor proteine transmembranare, membre ale familiei TLR, astfel aceste celule particip\u0103 at\u00e2t la distrugerea germenilor patogeni c\u00e2t \u015fi la celelalte procese, ca inflama\u0163ia, activarea imunit\u0103\u0163ii specifice, care stau la baza unor afec\u0163iuni precum lupus eritematos, pemfigus vulgar, psoriazis.
\u00cen patogenia lupusului eritematos sistemic s-a eviden\u0163iat implicarea masiv\u0103 a imunit\u0103\u0163ii \u00eennascute, astfel at\u00e2t acizii nucleici rezulta\u0163i din distruc\u0163ia celulara, c\u00e2t \u015fi complexele imune circulante pot activa receptroii TLR (TLR7 \u015fi TLR9) care se g\u0103sesc exprima\u0163i \u00een exces pe celulele mononucleate din sange. De asemenea, TLR 7 \u015fi TLR9 sunt implica\u0163i \u00een produc\u0163ia de autoanticorpi, iar TLR 2 \u015fi TLR 4 sunt implica\u0163i \u00een reglarea activit\u0103\u0163ii celulelor dendritice \u015fi a limfocitelor Th17, \u00een produc\u0163ia de IL17. \u00cen pemfigusul vulgar s-a eviden\u0163iat o cre\u015ftere a expresiei TLR4 la nivelul keratinocitelor, at\u00e2t \u00een stratul bazal c\u00e2t \u015fi \u00een straturile epidermice superioare. \u00cen psoriazis, keratinocitelele exprim\u0103 \u00een exces TLR1, TLR2 \u015fi TLR5, exist\u00e2nd diferen\u0163e semnificative \u00eentre pielea normal\u0103 \u015fi pielea lezional\u0103. Keratinocitul din pielea lezional\u0103, activat de receptrorii TLR (TLR2, TLR3, TLR4) cre\u015fte produc\u0163ia TNF alpha, IL8 \u015fi a altor citokine proinflamatoare. Activarea r\u0103spunsului imun \u00eenn\u0103scut \u015fi adaptativ, duce la activarea limfocitelor T, Th1 si \u00een special Th17, cu prodcerea \u00een exces a IL17. Citokinele proinflamatoare \u00een exces ca \u015fi ac\u0163iunea peptidei LL37 induce proliferarea excesiv\u0103 a keratinocitelor.

Innate immunity in autoimmune and inflammatory skin diseases<\/span> The innate immunity is one of the two aspects of general immunity, and it is also the first line of defense against pathogens. It is the immunity one is born with, it is fast, non-specific, without memory and independent of the nature of the antigen. The main mechanism of action consists in the expression on the surface of several cells (keratinocytes, NK cells, dendritic cells, monocytes, macrophages) of a toll-like protein family that recognize a molecular pattern on the surface of a microbial pathogen. Activation of these receptors induces a transduction signal that produces the activation of transcription factors (NFkB) with the production of TNF, IL1, IFN\u03b3 and ROS. These cytokines are directly or indirectly involved in pathogen destruction, inflammatory cell activation, induction of tissue restoration, mobilization and activation of the constituent cells of the adaptive immune system response. Keratinocytes, NK cells, dendritic cells, monocytes, macrophages express transmembrane proteins on their surface, which are members of the TLR family, so these cells participate in both the destruction of pathogenic germs and other processes, such as inflammation, activation of specific immunity, being the basis for diseases such as lupus erythematosus, pemphigus vulgaris, psoriasis.
In the pathogenesis of systemic lupus erythematosus, the massive implication of innate immunity has been demonstrated, so both nucleic acids resulting from cellular destruction and circulating immune complexes can activate TLR receptors (TLR7 and TLR9) that are found to be excessively expressed on mononuclear cells in the blood. Also, TLR 7 and TLR9 are involved in autoantibody production, and TLR 2 and TLR 4 are involved in regulating the activity of dendritic cells and Th17 lymphocytes in IL17 production. In pemphigus vulgaris, an increase in TLR4 expression in keratinocytes was seen in both the basal layer and the upper epidermal layers. In psoriasis, keratinocytes excessively express TLR1, TLR2 and TLR5, with significant differences between normal skin and lesional skin. The keratinocyte from injured skin, TLR receptor-activated (TLR2, TLR3, TLR4) increases the production of TNF alpha, IL8 and other proinflammatory cytokines. Activation of innate and adaptive immune response leads to the activation of T ymphocytes, Th1 and especially Th17 lymphocytes, with excess IL17 production. Excessive proinflammatory cytokines as well as the action of LL37 peptide induces excessive proliferation of keratinocytes.
Highlighting the implication of innate immunity in cutaneous pathology opens new perspectives on understanding the pathogenic processes that underlie several types of skin disorders, especially in the initiation phase of these processes.<\/p>\n
