Leziunile \u021besuturilor moi, ca de exemplu arsurile, ulcerele sau alte afec\u021biuni traumatice, reprezint\u0103 o grav\u0103 problem\u0103 de s\u0103n\u0103tate \u00een \u00eentreaga lume [1]. De\u0219i pentru tratarea acestor leziuni s-au ob\u021binut diferite materiale, problema unui tratament optim este departe de a fi rezolvat\u0103. Scopul acestui studiu a fost de a dezvolta noi hidrogeluri hidride, pe baz\u0103 de chitosan-acid hialuronic-bentonit\u0103. Chitosanul \u0219i acidul hialuronic sunt biopolimeri cu importante efecte antimicrobiene, hemostatice \u0219i cicatrizante [2, 3]. Bentonita are propriet\u0103\u021bi absorbante, dovedindu-se a fi ideal\u0103 pentru procesul de cicatrizare [4]. \u00cen matricea de hidrogel au fost \u00eencorporate arginina \u0219i betulina, cunoscute pentru efectele lor benefice \u00een procesul de cicatrizare [5, 6]. Prin varierea concentra\u021biei componentelor au fost ob\u021binute \u0219ase noi formul\u0103ri. Formul\u0103rile au fost caracterizate din punct de vedere morfologic, utiliz\u00e2nd microscopia electronic\u0103 de scanare, prin spectroscopie IR, grad de porozitate \u0219i umflare, studii de cinetic\u0103 \u0219i de stabilitate. Cele mai stabile formul\u0103ri au fost evaluate din punct de vedere biologic utiliz\u00e2nd teste in vitro (teste antimicrobiene \u0219i antioxidante) \u0219i in vivo (pe model de ran\u0103 indus\u0103 la \u0219obolanul diabetic). Cea mai bun\u0103 formulare s-a dovedit a fi hidrogelul chitosan-acid hialuronic-bentonit\u0103 (1,8:0,5:4,5) cu un con\u021binut de arginin\u0103 \u0219i betulin\u0103 de 1%. Aceast\u0103 formulare a prezentat caracteristici fizico-chimice \u0219i reologice \u00eembun\u0103t\u0103\u021bite. Studiile de cinetic\u0103 au eviden\u021biat faptul c\u0103 substan\u021bele active (arginina\/betulina) sunt eliberate \u00een propor\u021bie de 70% \u00een decurs de 6 ore, ceea ce \u00eenseamn\u0103 c\u0103 formularea are o ac\u021biune de lung\u0103 durat\u0103. \u00cen plus, aceast\u0103 formulare a prezentat efecte antimicrobiene, antioxidante \u0219i cicatrizante \u00eembun\u0103t\u0103\u021bite.<\/p>\n

The healing potential of new chitosan-hyaluronic acid based hydrogels<\/span>

The soft tissue injuries, such as burn, ulcer or other traumatic damages, represent a major health care problem in the entire world [1]. Although several wound dressings materials have been developed, the problem of the wounds management is far from being solved. The aim of this study was to develop new hybrid hydrogels based on chitosan-hyaluronic acid-bentonite. Chitosan and hyaluronic acid are biopolymers which mimic the properties of the extracellular matrix and have important biological effects such as antimicrobial, haemostatic and wound healing [2, 3]. Bentonite is an absorbent natural clay which has ideal properties for wound healing [4]. Into hydrogel matrix, arginine and betulin, known for their important biological effects for healing process, have been incorporated [4, 5]. Varying the concentration of components, six new innovative formulations have been developed. The formulations were characterized in terms of morphology using scanning electron microscopy, IR spectroscopy, swelling and porosity degree, kinetic studies and stability properties. The most proper and stable formulations were biologically evaluated using in vitro (antimicrobial, antioxidant assays) and in vivo tests (using wound model induced to diabetic rat). The best formulation was the chitosan:hyaluronic acid: bentonite (1.8:0.5:4.5) hydrogel containing arginine and betulin in concentration of 1%. This formulation showed improved physico-chemical and rheological characteristics. The kinetic study revealed that the active substances (arginine\/betulin) is released during 6 h in ratio of 70%, which means this formulation remain active long time. Moreover this formulation showed improved antimicrobial, antioxidant and healing effects.<\/p>\n

Scurt CV<\/span><\/p>\n

Dr. Lenuta Profire is Professor of Pharmaceutical Chemistry, Faculty of Pharmacy, \u201cGrigore T. Popa\u201d University of Medicine and Pharmacy, Iasi, Romania. She received her PhD in 1991 with the thesis Research concerning synthesis of new bioactive xanthine derivatives. Since 2016 she is Dean of Faculty of Pharmacy. She is member of the scientific and professional organisations and managed several national and international research projects. Other important achievements: PhD supervisor (since 2008), Director of Doctoral School (2012-2016), Member of Doctoral School Council (2012-present), Member of the National Council for Attesting University Titles, Diplomas and Certificates (2011-2012, 2016-present).
Research Interests:<\/strong>
Improving the pharmacological and pharmacokinetic profile of the biological active compounds and drugs using biopolymers as delivery systems (based on chitosan, chitosan\/hyaluronic acid, chitosan\/xanthan, cellulose\/chondroitin sulphate). Design and characterization of new biological active compounds focused on new heterocycle derivatives (based on xanthine, thiazolidine-4-one, pyrazolin-5-one, azetidin-2-one, aminoacids scaffold) and their biological evaluation (antimicrobial, antioxidant, anti-inflammatory, antidiabetes effects).<\/p>\n
