Introducere. Celulele prezentatoare de antigen din piele sunt \u00een principal celule Langerhans (LCs) care sus\u021bin func\u021bii imune \u00een cadrul re\u021belei celulare cutanate, aceste celule fiind considerate santinele imune majore. Dup\u0103 activare LCs migreaz\u0103 \u00een ganglionii limfatici activ\u00e2nd celulele imune adaptative (ex. limfocitele T \u0219i B), care ulterior vor dezvolta o multitudine de tipuri de r\u0103spunsuri imune. C\u00e2nd mecanismele imune reglatoare nu sunt eficiente, LCs devin implicate \u00een diverse patologii cutanate, de la boli autoimune la cancer.
Materiale \u0219i metode. Utiliz\u00e2nd protocoale standard de imunohistochimie, biopsii tisulare au fost investigate pentru prezen\u021ba \u0219i distribu\u021bia LCs.
Rezultate. \u00cen psoriazis markerul CD1a exprimat de LCs, este o molecul\u0103 implicat\u0103 \u00een accelerarea proces\u0103rii antigenului. C\u00e2nd CD1a este supra-exprimat \u00een leziunile psoriatice, simptomatologia este mai intens\u0103. \u00cen biopsiile leziunilor psoriatice, un num\u0103r crescut de LCs epidermale au fost g\u0103site \u00een compartimentele bazale\/suprabazale. LCs au fost identificate \u00een melanoamele cutanate sus\u021bin\u00e2nd r\u0103spunsul imun local. Pe l\u00e2ng\u0103 limfocitele T care infiltreaz\u0103 tumora, LCs, macrofagele, celulele dendritice \u0219i celulele natural uciga\u0219e constituie armata imun\u0103\u021bii locale. Eficien\u021ba terapiilor imune este determinat\u0103 de componen\u021ba celular\u0103 a micromediului tumoral \u0219i de interac\u021bia dintre celule imune \u0219i celule tumorale. \u00cen timp ce o serie de melanoame abund\u0103 \u00een LCs, altele sunt lipsite de acest tip celular. Concluzie. Caracteristicile moleculare ale LCs determin\u0103 capacitatea acestora de a infiltra micromediul tumoral, cum ar fi expresia diferen\u021bial\u0103 a Toll-like Receptors de pe suprafa\u021ba LCs.

Introduction. Skin\u2019s antigen presenting cells are mainly Langerhans cells (LCs) that within their created cellular network sustain immune functions, being considered the main immune sentinels of the skin. LCs upon activation can migrate to skin draining lymph nodes priming adaptive immune cells (e.g. T- and B-lymphocytes), developing further a broad range of immune responses. When immune regulatory mechanisms fail, LCs become involved in various skin\u2019s pathology, ranging from autoimmunity to cancer.
Materials and methods. Tissue biopsies were investigated for the presence and distribution of LCs using standard immunohistochemistry protocols.
Results. In psoriasis the CD1a marker expressed on LCs, is a molecule involved in the active antigen-processing events. When CD1a is over-expressed, psoriatic symptoms are intensified. In our investigated skin psoriatic biopsies, increased numbers of epidermal LCs were found located predominantly in the basal\/suprabasal compartment. LCs were found in skin melanoma tissues where they participate at the local immune response. Besides T cells that infiltrate, LCs, macrophages, dendritic and natural killer cells constitute the local immune army. The efficacy of immune therapies resides in the cellular composition of the tumor immuno-microenvironment, and on the crosstalk between immune and tumor cells. While some skin melanomas have high amounts of LCs, others lack these cells. Conclusion. The molecular characteristics of LCs determine their ability to infiltrate the tumor microenvironment, such as the expression of different Toll-like Receptors on the surface of LCs.

Dup\u0103 activare LCs migreaz\u0103 \u00een ganglionii limfatici activ\u00e2nd celulele imune adaptative (ex. limfocitele T \u0219i B), care ulterior vor dezvolta o multitudine de tipuri de r\u0103spunsuri imune. C\u00e2nd mecanismele imune reglatoare nu sunt eficiente, LCs devin implicate \u00een diverse patologii cutanate, de la boli autoimune la cancer.
Materiale \u0219i metode. Utiliz\u00e2nd protocoale standard de imunohistochimie, biopsii tisulare au fost investigate pentru prezen\u021ba \u0219i distribu\u021bia LCs.
Rezultate. \u00cen psoriazis markerul CD1a exprimat de LCs, este o molecul\u0103 implicat\u0103 \u00een accelerarea proces\u0103rii antigenului. C\u00e2nd CD1a este supra-exprimat \u00een leziunile psoriatice, simptomatologia este mai intens\u0103. \u00cen biopsiile leziunilor psoriatice, un num\u0103r crescut de LCs epidermale au fost g\u0103site \u00een compartimentele bazale\/suprabazale. LCs au fost identificate \u00een melanoamele cutanate sus\u021bin\u00e2nd r\u0103spunsul imun local. Pe l\u00e2ng\u0103 limfocitele T care infiltreaz\u0103 tumora, LCs, macrofagele, celulele dendritice \u0219i celulele natural uciga\u0219e constituie armata imun\u0103\u021bii locale. Eficien\u021ba terapiilor imune este determinat\u0103 de componen\u021ba celular\u0103 a micromediului tumoral \u0219i de interac\u021bia dintre celule imune \u0219i celule tumorale. \u00cen timp ce o serie de melanoame abund\u0103 \u00een LCs, altele sunt lipsite de acest tip celular. Concluzie. Introduction. Skin\u2019s antigen presenting cells are mainly Langerhans cells (LCs) that within their created cellular network sustain immune functions, being considered the main immune sentinels of the skin. LCs upon activation can migrate to skin draining lymph nodes priming adaptive immune cells (e.g. T- and B-lymphocytes), developing further a broad range of immune responses. When immune regulatory mechanisms fail, LCs become involved in various skin\u2019s pathology, ranging from autoimmunity to cancer.
Materials and methods. Tissue biopsies were investigated for the presence and distribution of LCs using standard immunohistochemistry protocols.
Results. In psoriasis the CD1a marker expressed on LCs, is a molecule involved in the active antigen-processing events. When CD1a is over-expressed, psoriatic symptoms are intensified. In our investigated skin psoriatic biopsies, increased numbers of epidermal LCs were found located predominantly in the basal\/suprabasal compartment. LCs were found in skin melanoma tissues where they participate at the local immune response. Besides T cells that infiltrate, LCs, macrophages, dendritic and natural killer cells constitute the local immune army. The efficacy of immune therapies resides in the cellular composition of the tumor immuno-microenvironment, and on the crosstalk between immune and tumor cells. While some skin melanomas have high amounts of LCs, others lack these cells. Conclusion. The molecular characteristics of LCs determine their ability to infiltrate the tumor microenvironment, such as the expression of different Toll-like Receptors on the surface of LCs.

Materiale \u0219i metode. Utiliz\u00e2nd protocoale standard de imunohistochimie, biopsii tisulare au fost investigate pentru prezen\u021ba \u0219i distribu\u021bia LCs.
Rezultate. \u00cen psoriazis markerul CD1a exprimat de LCs, este o molecul\u0103 implicat\u0103 \u00een accelerarea proces\u0103rii antigenului. C\u00e2nd CD1a este supra-exprimat \u00een leziunile psoriatice, simptomatologia este mai intens\u0103. \u00cen biopsiile leziunilor psoriatice, un num\u0103r crescut de LCs epidermale au fost g\u0103site \u00een compartimentele bazale\/suprabazale. LCs au fost identificate \u00een melanoamele cutanate sus\u021bin\u00e2nd r\u0103spunsul imun local. Pe l\u00e2ng\u0103 limfocitele T care infiltreaz\u0103 tumora, LCs, macrofagele, celulele dendritice \u0219i celulele natural uciga\u0219e constituie armata imun\u0103\u021bii locale. Eficien\u021ba terapiilor imune este determinat\u0103 de componen\u021ba celular\u0103 a micromediului tumoral \u0219i de interac\u021bia dintre celule imune \u0219i celule tumorale. \u00cen timp ce o serie de melanoame abund\u0103 \u00een LCs, altele sunt lipsite de acest tip celular. Concluzie. Caracteristicile moleculare ale LCs determin\u0103 capacitatea acestora de a infiltra micromediul tumoral, cum ar fi expresia diferen\u021bial\u0103 a Toll-like Receptors de pe suprafa\u021ba LCs.

Materials and methods. Tissue biopsies were investigated for the presence and distribution of LCs using standard immunohistochemistry protocols.
Results. In psoriasis the CD1a marker expressed on LCs, is a molecule involved in the active antigen-processing events. When CD1a is over-expressed, psoriatic symptoms are intensified. In our investigated skin psoriatic biopsies, increased numbers of epidermal LCs were found located predominantly in the basal\/suprabasal compartment. LCs were found in skin melanoma tissues where they participate at the local immune response. Besides T cells that infiltrate, LCs, macrophages, dendritic and natural killer cells constitute the local immune army. The efficacy of immune therapies resides in the cellular composition of the tumor immuno-microenvironment, and on the crosstalk between immune and tumor cells. While some skin melanomas have high amounts of LCs, others lack these cells. Conclusion. The molecular characteristics of LCs determine their ability to infiltrate the tumor microenvironment, such as the expression of different Toll-like Receptors on the surface of LCs.
